Cardiovascular diseases (CVD) accounts for a majority of death worldwide, thereby, numerous of relevant drugs for CVD are under development. However, drug development has always been considered as a time-consuming and costly process. By improving the reliability and accuracy on drug assessments, it is possible to accelerate the process on developing a new drug. It is proved that maturation of cardiomyocytes plays a main role in drug screening, furthermore, current researches on maturation of cardiomyocytes have become a hit, among them, conductive material and topography method gain a lot attention. In this research, conductive material silver nano wires (AgNWs) and 800 nm wide grooves are combined to be the culture substrate for cardiomyocytes in vitro (NRVM), subsequently, a polymer cantilever is fabricated with this combined material. After seeding cardiomyocytes on the surface of this AgNWs-embedded nano-patterned PDMS cantilever, cell beating behaviors are observed. On the one hand, by embedding most part of AgNWs into PDMS, the adhesion between AgNWs with PDMS is greatly improved, and the effective protection for AgNWs from oxidation is provided. On the other hand, with embedded structure, cell alignment is completely achieved without the disorders caused by AgNWs, in the meanwhile, AgNWs-embedded PDMS cantilever shows better cell-to-cell connections which guarantees its further application. Moreover, with an enhancement on maturation of cardiomyocytes, this cantilever can offer a more accurate information on drug testing.
